Colorectal and endometrial cancers have many features in common. Both have identifiable histopathologic precursor lesions. Histopathologic progression from adenoma to carcinoma in colon cancer also is seen in the atypical hyperplasia endometrial carcinoma sequence. Colorectal and endometrioid adenocarcinoma of the uterus also have similar histologic appearances. Certain alterations in oncogenes and tumor supressor genes, such as p53 and K-ras, in colorectal cancer also are seen in endometrial cancer.18, 19 Microsatellite instability in colorectal cancer20 also has been found in endometrial cancer.11 In women of families afflicted with hereditary nonpolyposis colorectal cancer of the Lynch syndrome variant, endometrial carcinoma is the most common noncolorectal malignancy reported.21 Based on those similarities, we postulated that nonsteroidal antiinflammatory drugs also inhibit growth of an endometrial carcinoma cell line in way a similar to that described for colorectal cancer. Our study shows a dosedependent inhibition of Ishikawa cell growth by ASA in the range of 15 mM. Those results compare favorably to data of Elder et al4 who examined exposure of colon adenocarcinoma cell lines to the same concentrations of sodium salicylate. Several mechanisms of growth inhibition by nonsteroidal anti-inflammatory drugs on colon cancer cell lines have been suggested, including reduction of the rate of cellular proliferation and induction of programmed cell death, or apoptosis.3, 4 In this study, the growth inhibitory effect of ASA seems to be mediated in part by induction of apoptosis. The gene product of bcl-2, originally described in non-Hodgkin lymphomas, 22 enhances cell survival by inhibiting apoptosis.23 Several studies found bcl-2 in normal endometrium.24, 25 Bcl-2 expression in glandular cells peaks at the end of the follicular phase and disappears in the late secretory phase. The effect of bcl-2 in this setting might be prevention of apoptosis during the proliferative phase. In malignant endometrium, bcl-2 expression was highly correlated to hormone receptors and tumor differentiaAspirin Effects on Endometrial Cancer Cell Growth 425.

Et al., 2000a, b; Lowe et al., 2002; Baker et al., 2003b; Joseph et al., 2004 ; . In this study, the agonist actions of adrenaline and noradrenaline were inhibited in a very similar manner to that of isoprenaline and are therefore acting through the catecholamine site of the human 1-adrenoceptor. This is in agreement with previous findings in the ferret Lowe et al., 2002 ; . All of the 12 antagonists used in this study have higher affinity KD value ; for the catecholamine site than the secondary site Table 1 ; . This is also true for CGP 12177 itself and carvedilol which are neutral antagonists of the catecholamine site and agonists of the secondary site ; and alprenolol and pindolol which are agonists of both sites; Pak and Fishman, 1996; Konkar et al., 2000b; Lowe et al., 2002; Baker et al., 2003b ; . Thus, regardless of the efficacy of the ligand, no ligand has yet been described that is able to bind to the secondary site with greater affinity. However, the rank order of antagonist affinities at the catecholamine site which is identical with isoprenaline, adrenaline, and noradrenaline ; is markedly different from the rank order of affinities at the secondary site Table 1 and Fig. 6 where the poor correlation is clearly demonstrated ; . ICI 118551, bupranolol, and nadolol all have higher than expected affinity, and atenolol has a lower than expected affinity for the secondary site than if it was just a low-affinity mirror of the catecholamine site. This is similar to findings recently reported by Joseph et al. 2004 ; also at the human 1-adrenoceptor, but there appear to be some differences between species cf. Lowe et al., 2002 in the ferret heart, this report and Joseph et al., 2004 using the human 1-adrenoceptor ; . Thus, as well as the differences in ligand efficacy at the two sites, the different ligand affinities also suggest that the secondary site is indeed a separate pharmacological entity. Cimaterol stimulated responses that were inhibited by all antagonists to yield much higher affinity values than those achieved with CGP 12177, suggesting that cimaterol is not acting via the secondary site. However, all of the KD values obtained showed a consistently higher affinity than when the catecholamines were present. This raises the possibility that cimaterol is acting through a third, higher-affinity conformation of the 1-adrenoceptor and that many ligand-dependent states of the 1-adrenoceptor may exist Molenaar, 2003; Arch 2004; Fig. 6 ; . The rank order of antagonist affinity Table 1; Fig. 6 ; , however, is the same for cimaterol as for the catecholamines, suggesting that cimaterol is likely to stimulate the 1-adrenoceptor via the catecholamine site. Cimaterol also behaved exactly the same as the catecholamines in the presence of CGP 12177 Fig. 3 ; . Here, CGP 12177 and naltrexone.

The cells which are inhibited by HLA-G bearing DC carry specific receptors for HLA-G. There are, for example, receptors on NK cells for HLA-G namely KIR2DL4, ILT2 and ILT4 that down regulate cell proliferation upon interaction with HLA-G.25, 26 The presence of HLA-G receptors on CD4 T cells has also been shown for immunoglobulin like transcript 2 ILT2 ; indicating this molecule a possible candidate mediating inhibitory effects.27 In the present study, the inhibition of T cells proliferation was observed only when cells were treated with IFN- Table 3 ; . Thus IFN- and HLA-G molecules on the dendritic cells are two components contributing to this phenomenon, limiting cells proliferation, thereby reducing the potential of T-cells. It is quite evident that the cells bearing HLA-G are an important negative immuneregulatory constituent for this suppressing effect. It is speculated that IFN- by increasing HLA-G on dendritic cells can also down regulate inflammatory response in vivo mediated by T cells in islet tissues of pancreatic gland. It has been reported that higher HLAG levels correlated with a better outcome of heart transplantation.28 Levels of HLA-G on dendritic cells may also be correlated to disease activity in diabetic patients and it could also serve as a useful marker for disease progress and treatment. We would like to propose that HLA-G may be a factor limiting autoimmune response, thus potentially constituting a new component for controlling and suppressing the detrimental role of T-cells in the pathogenesis of diabetes type 1.
